Novel Lupane Triterpenoid-Indazole Conjugates with Oxime Linkage

Betulin 1 and its derivatives demonstrate wide range of biological activity. On the other hand indazole
skeleton and oxime linkage are attractive, pharmaceutically active structural scaffolds in medicinal
chemistry. Thus, introduction of indazole skeleton and oxime functionality into the structure of betulin
is reasonable approach to potential cytotoxic agents.
Synthesis of the target compounds 2 was started with oxidation of betulin 1. The obtained betulonic acid
3 was successfully purified through formation of its cyclohexylammonium salt. Lupane triterpenoidindazole
hybrids 2 with oxime linkage were obtained from betulonic acid chloride 4 and corresponding
oximes 5 with medium to high yields.
